SISTEM PENGHANTARAN OBAT BERBASIS NANOKARIER UNTUK OPTIMASI BIOAVAILABILITAS OBAT ORAL: TINJAUAN SISTEMATIS DAN ANALISIS BIBLIOMETRIK

Latar Belakang: Pemberian obat secara oral adalah cara paling umum untuk menyediakan obat-obatan, namun banyak obat yang memiliki bioavailabilitas rendah karena tidak larut dengan baik, tidak mudah melewati usus, terurai di perut, dan melewati metabolisme terlebih dahulu. Perangkat pengiriman obat berbasis nanoteknologi telah muncul sebagai pendekatan yang layak untuk mengatasi kendala farmakokinetik ini. Tujuan: Penelitian ini bertujuan untuk mengkaji tren penelitian, kerangka pengetahuan, dan kemajuan teoritis sistem pengiriman obat berbasis nanoteknologi dalam meningkatkan ketersediaan hayati obat-obatan yang diberikan secara oral. Kami melakukan tinjauan pustaka sistematis (SLR) dan analisis bibliometrik menggunakan metodologi PRISMA. Pencarian literatur dilakukan secara komprehensif melalui dua sumber metadata, yaitu ScienceDirect dan Publish or Perish, masing-masing menghasilkan 1.000 artikel dengan rentang waktu 2016–2026, sehingga total 2.000 artikel diidentifikasi. Sebelum penyaringan PRISMA, seluruh 2.000 artikel dianalisis menggunakan VOSviewer untuk memetakan gap penelitian dan novelty melalui visualisasi jaringan kata kunci. Setelah proses penyaringan berdasarkan kriteria inklusi dan eksklusi, diperoleh 30 artikel terpilih (n = 30). Studi ini mengungkapkan bahwa nanopartikel, ketersediaan hayati, pemberian obat oral, dan metode pemberian obat merupakan masalah penelitian yang dominan. Penelitian saat ini menggunakan platform nanocarrier, termasuk nanopartikel lipid padat, liposom, dan nanopartikel silika mesopori, semakin difokuskan pada peningkatan kelarutan, permeabilitas, dan regulasi pelepasan obat. Kesimpulan: Metode pemberian obat berdasarkan nanoteknologi dapat membantu membuat obat oral lebih tersedia secara hayati. Ini juga merupakan cara potensial untuk menyiasati formulasi sistem penghambat obat oral tradisional.
