Efektivitas Propranolol Pada Terapi Post-Traumatic Stress Disorder (Ptsd) : Systematic Review

Post-traumatic stress disorder (PTSD) merupakan gangguan psikiatrik kronis yang ditandai dengan memori traumatis yang kuat dan sulit dimodifikasi. Propranolol, melalui mekanisme blokade β-adrenergik, diduga dapat menurunkan intensitas emosional memori traumatis sehingga menyebabkan penurunan gejala PTSD. Systematic review ini bertujuan mengevaluasi efektivitas propranolol sebagai terapi berbasis rekonsolidasi memori dalam menurunkan gejala PTSD pada pasien dewasa. Pencarian literatur dilakukan melalui PubMed dan ScienceDirect untuk periode 1 Januari 2015 – 20 November 2025 menggunakan kata kunci ”Post traumatic stress disorder” OR ” PTSD” AND ”Propranolol”. Seleksi studi dilakukan berdasarkan kriteria PICOS, dengan inklusi terbatas pada randomized controlled trial (RCT). Kualitas metodologis dinilai menggunakan The Revised JBI Critical Appraisal Checklist for RCTs, dan analisis dilakukan secara deskriptif kualitatif. Dari 164 artikel yang teridentifikasi, empat RCT memenuhi kriteria inklusi. Seluruh studi melaporkan penurunan gejala PTSD pada kelompok propranolol maupun kelompok pembanding. Namun, hanya satu dari empat RCT yang menunjukkan penurunan gejala PTSD yang secara signifikan lebih besar pada kelompok propranolol. Bukti yang tersedia menunjukkan bahwa efektivitas propranolol sebagai terapi berbasis rekonsolidasi memori pada PTSD masih terbatas dan tidak konsisten. Oleh karena itu, propranolol belum dapat disimpulkan efektif secara klinis namun tetap berpotensi sebagai adjuvan eksperimental. Untuk itu penelitian lanjutan dengan protokol reaktivasi memori yang lebih terstandarisasi serta desain yang lebih kuat masih diperlukan untuk memastikan potensi perannya dalam tatalaksana PTSD.
